2.1 人脂肪源性干细胞的生物学特性鉴定结果 成功分离培养的人脂肪源性干细胞表现出间充质干细胞的典型生物学特性。在标准培养条件下,细胞呈梭形或多角形,增殖性强,可稳定传代(图2A)。流式细胞术分析结果证实,所培养的细胞符合国际细胞治疗协会(ISCT)的标准[11]:细胞高表达间充质干细胞标志物CD44 (97.84±1.25)%、CD90 (99.43±0.52)%及CD105 (98.74±0.86)%,同时几乎不表达造血干细胞标志物CD34 (0.15±0.08)%、CD45 (0.23±0.11)%和HLA-DR (0.14±0.07)% (图2B)。此外,人脂肪源性干细胞表现出明确的多向分化潜能[11]。经成骨诱导2周后,碱性磷酸酶染色呈强阳性(图2C);经成脂诱导2周后,油红O染色可见大量胞内脂滴形成(图2D)。综上所述,成功获得并鉴定了一批高纯度、具有多向分化能力的人脂肪源性干细胞,为后续的神经定向分化实验提供了可靠的细胞来源。 "
2.2 第一阶段诱导将人脂肪源性干细胞转分化为神经祖细胞 在第一阶段诱导过程中,人脂肪源性干细胞的形态发生了显著的神经谱系转变。诱导4 d后,细胞逐渐由梭形变为紧凑的聚集形态,并形成了典型的“神经花瓣”状结构[13](图3A)。 与形态学变化一致,分子水平检测也证实了细胞向神经祖细胞的成功转化。Western blot和RT-qPCR分析显示,神经祖细胞关键标志物PAX6和SOX1的蛋白与mRNA表达水平均从诱导第2天起显著上调,并在第4天达到峰值(P < 0.001),随后维持在较高水平(图3B,C)。 为精准评估转分化效率,免疫荧光染色结果显示,在诱导第4天,细胞群体已高表达PAX6和SOX1,且蛋白主要定位于细胞核。定量分析表明,PAX6阳性细胞率高达(85.33±6.27)%,SOX1阳性细胞率为(83.12±5.81)%(图3D)。 综上,形态学、分子生物学及细胞化学证据共同表明,Dorsomorphin、CHIR99021和SB431542组合能够将人脂肪源性干细胞诱导为神经祖细胞。 "
2.3 第二阶段诱导促使神经祖细胞成功分化为运动神经元样细胞 进入第二阶段诱导后,神经祖细胞逐渐分化为成熟的神经元形态[14]。至第12天,细胞胞体变圆或呈多角形,并伸出长度可达胞体直径5-10倍的复杂神经突起,部分细胞间形成初步的网络连接(图4A)。 分子水平的表达变化与形态学转变同步。Western blot和RT-qPCR分析均显示,运动神经元标志物(微管蛋白β3、ISL1、HB9和胆碱乙酰转移酶)的蛋白与mRNA表达水平在诱导过程中显著上调,并于第12天达到表达高峰(P < 0.001),随后维持在较高水平(图4B,C)。 为进一步验证分化效率与特异性,于诱导第12天进行了免疫荧光双染(图4D),结果显示,细胞群体中高比例共表达关键运动神经元标志物:(78.56±7.32)%的细胞共表达微管蛋白β3和ISL1,同时(75.04±6.81)%的细胞共表达HB9和胆碱乙酰转移酶。这些数据表明,第二阶段的CHIR99021、维甲酸和Purmorphamine组合能够特异性驱动神经祖细胞向运动神经元样细胞高效分化。 "
