Abstract:

背景:正畸矫治器的戴用破坏了牙齿及其周围组织的环境,造成菌斑堆积,引起一系列不良反应。
目的:检测含铜抗菌不锈钢正畸矫治器的细胞毒性。
方法:①CCK-8检测细胞增殖:取生长旺盛的人口腔上皮癌细胞KB,实验组加入含铜抗菌不锈钢材料浸提液,对照组加入普通医用不锈钢材料浸提液,阴性对照组加入DMEM培养基,阳性对照组加入苯酚,分别培养24,48,72 h。②流式细胞仪Annexin V-FITC/PI双染法检测细胞凋亡:观察普通医用不锈钢、含铜抗菌不锈钢材料对数生长期末人口腔上皮癌细胞KB凋亡的影响,设置空白对照组。
结果与结论:①培养48 h后,阳性对照组A值随作用时间延长逐渐降低,其余组细胞A值随时间延长逐渐递增(P < 0.05),且实验组高于对照组(P < 0.05)。普通不锈钢和抗菌不锈钢材料的细胞毒性均为1级。②普通医用不锈钢、含铜抗菌不锈钢组凋亡细胞均较少,且含铜抗菌不锈钢组少于普通医用不锈钢组(P均< 0.05)。表明含铜抗菌不锈钢材料生物相容性良好,毒性等级与医用不锈钢相同,符合口腔正畸材料的临床应用要求。
